An International Summit Addressing ADC Safety Assessment and Clinical Translation
The ADC Toxicity Summit is an international conference that brings together experts in toxicology, safety assessment, translational research, preclinical development, and clinical medicine who are involved in the development of antibody-drug conjugates, or ADCs. Participants discuss strategies for predicting, preventing, and mitigating ADC-related toxicities.
The fourth edition of the Summit is expected to bring together more than 70 toxicologists, pharmacologists, translational scientists, clinicians, and safety leaders. The program will address critical safety challenges in ADC development, including interstitial lung disease, or ILD, ocular toxicity, neurotoxicity, and target-independent toxicity.
The Summit will also explore approaches to improving the predictive performance and clinical translatability of preclinical safety assessments. Topics will include advanced in vitro and in vivo models, including organoids, translational biomarkers, computational toxicology, pharmacokinetic and pharmacodynamic analyses, and New Approach Methodologies, or NAMs, promoted by the U.S. Food and Drug Administration.
HiLung’s Role at the Summit
Rather than participating as a pharmaceutical company developing ADC products, HiLung will join the Summit as a technology partner supporting the translation of preclinical safety findings generated using human respiratory tissues into a better understanding of clinically observed lung injury.
Predicting the risk of drug-induced lung injury, particularly ILD, before clinical trials remains a major challenge in ADC development. Key themes at the Summit will include the mechanisms underlying ADC-associated pulmonary toxicity, the responses of alveolar epithelial cells, macrophages, and fibroblasts, and the early detection and clinical management of ILD.
Based on its proprietary technologies for generating human induced pluripotent stem cell-derived alveolar and airway organoids, HiLung provides in vitro models that reproduce multicellular interactions within the human respiratory system. These models are available through contract research services and as ready-to-use cell and tissue plates. They are intended to address one of the major challenges in drug development: the difficulty of predicting pulmonary toxicities, including interstitial lung disease.
Using HiLung’s alveolar organoid platform, HiFOAms™, and its human iPSC-derived tissue-resident alveolar macrophages, HiTRAM™, research is underway to evaluate multiple aspects of pulmonary responses to ADCs, free payloads, and related compounds using human cell-based systems. These assessments include alveolar epithelial injury, macrophage responses, inflammatory reactions, and fibroblast activation.
By combining histopathological assessment, cell death and injury assays, cytokine profiling, gene expression analysis, and single-cell RNA sequencing, HiLung supports the investigation not only of whether cytotoxicity occurs, but also of the specific cell populations involved, the temporal progression of lung injury, and the underlying mechanisms of toxicity.
Contributing to the Development of Safer ADCs
Through its participation in the Summit, HiLung aims to strengthen its collaboration with ADC developers, toxicologists, clinicians, and regulatory science experts. The company will continue working toward the standardization of pulmonary toxicity assessment using human respiratory models and the improvement of clinical translatability.
HiLung will continue integrating human iPSC technology, organoid engineering, and translational science to support the implementation of respiratory NAMs that complement animal studies and contribute to the development of safer and more effective ADCs and other emerging therapeutic modalities.
